When youre pregnant, you usually have to go nine months or longer without using certain active ingredients in your skin-care routine (farewell: dear, retinol). But just because a few ingredients arent recommended to be used in your regimen for a while doesnt mean that skin care has to fall by the waysidethere are plenty of barrier-boosting actives that are A-okay to use when youre expecting. For some guidance, a celebrity esthetician is sharing her entire pregnancy skin-care regimen.
Overall, Natalie Aguilar, celebrity facialist and dermatological nurse, had to swap out six of her daily skin-care staples when she got pregnant. My skin is very resilient, but easily pigments, so preventing facial discoloration and pigmentation has been my main goal before and during my pregnancy, she says. To do the brightening job during her pregnancy, Aguilar nixed her typical retinol serum, salicylic acid cleanser, stem cell moisturizer, and chemical SPF, but has effective substitutes in their place (like kojic acid, for one).
Now, Aguilars regimen still contains some complexion-evening ingredients, but shes also incorporating skin-soothing and hydrating products to balance her finicky skin. During pregnancy, our skin may experience extreme dryness, sensitivity, and even melasma that we didnt have before thanks to the shift of hormones, she says. Pre-pregnancy, Aguilar turned to a salicylic acid-spiked cleanser for sloughing off dead skin cells. My new substitute is this PCA Skin Pigment Bar, which uses natural skin-brightening and exfoliating ingredients such as kojic acid, which is derived from mushrooms or fermented rice, and azelaic acid, which is a naturally occurring acid found in grains, she says.
Aguilar had to change her go-to toner when she got pregnant, since her active ingredient-packed product started to burn and sting her skin. I swapped for this toning essence, which is alcohol-free and has skin-soothing ingredients like chamomile and rose extracts, she says.
Though Aguilar always used a vitamin B3 and vitamin C serum daily, she found that her skin no longer tolerated them when used together. Instead, I use this vitamin B3 serum during the day, and a vitamin C serum at night. The active ingredient, vitamin B3 (also known as niacinamide) does everything from hydrate to smooth and even out the complexion.
To aid in her skin-brightening goals, Aguilar uses a vitamin C serum every day. Though the one in her cabinet is by PCA Skin, this serum by Ghost Democracy uses a stable vitamin C derivative that works for all skin types, plus it contains bakuchiol, a plant-based retinol alternative.
I added a hyaluronic acid [to my routine] as my skin began to suffer extreme dehydration, no matter how much water I was drinking, says Aguilar. Hyaluronic acid is a safe, water-binding serum, and I began to use one twice daily and instantly felt relief. This drugstore find will give your skin moisturizing, fine line-plumping benefits, and works for the most sensitive of skin types.
My stem cell moisturizer and eye cream were swapped out, says Aguilar, noting that though there isnt much research on the safety of using topical stem cells during pregnancy, its best to avoid products you just arent sure about. Instead, she turns to peptides, which are collagen-stimulating proteins that are naturally found in your skin. I switched to using a rich, hydrating cream that contains plant peptides, which are proteins that help strengthen the skins barrier.
To stay on the safe side in her beauty regimen, Aguilar swapped her chemical-based sunscreen for this mineral-based option. This is safe enough to be used on babies and children, she says. I feel safer using a mineral sunscreen that provides a protective physical shield and that doesnt penetrate the skin. Her tip? Reapply two to three times a day for ultimate sun protection.

Teaming up with blogger Mehreen Baig, Mosley explored the freaky world of lip and nose fillers, microneedling and botox, and bravely volunteered to have his own crows feet blitzed by a gadget which, as its operator enthused, melts the skin instantaneously. Once the rawness and swelling on his face had subsided, Mosley was disgruntled to find that it hadnt made much difference.
Other customers were left similarly deflated. Julie, whose fractionated CO2 laser treatment left her face covered in tatters of dead skin, enjoyed some improved skin elasticity, but tests revealed no noticeable dermatological changes. The only treatment that seemed to have a significant effect was the stem-cell facelift undergone by Kim, who paid 6,000 for the privilege of having the cells injected into her cheekbones. She was delighted with her smoother, younger-looking face.
Mosley had assembled a panel of punters to look at before and after photos and assess whether the treatments had made the contestants look more attractive. They lost their personality, one man said. As dermatologist Tamara Griffiths warned, then, its a case of buyer beware.

I think that the gold standard will eventually be finding ways to regenerate and kick-start our own collagen instead of doing a facelift. Kellett is already trying out cutting-edge technology to accomplish this, such as a laser that delivers growth factors right in the dermis to regenerate tissue. I think thats the future of aging, says Kellett, who is just about to launch such a treatment at her clinic. The pill wouldnt deliver all the benefits of physical activity, such as the positive impact on our mood, but it would replicate its effects on our body. It wont take the place of walking around outside and soaking up natureit cant do the mental part of it. But as far as the physiologic, biochemical part of it, were really understanding that better and making big strides. Its exciting.

Steadman Philippon Research Institute (SPRI) Receives Multi-Million-Dollar Matching Grant from the National Institutes of Health – GlobeNewswire
VAIL, Colorado, Aug. 20, 2020 (GLOBE NEWSWIRE) -- Steadman Philippon Research Institute (SPRI) has been granted the prestigious Regenerative Medicine Innovation Project Investigator-Initiated Clinical Trials award from the National Institutes of Health (NIH). SPRI Chief Scientific Officer Johnny Huard, Ph.D. will serve as the principal investigator. Marc J. Philippon, M.D., who serves as managing partner of The Steadman Clinic and co-chair of SPRI and Scott Tashman, Ph.D., director of biomedical engineering at SPRI, will serve as co-principal investigators. The clinical trials are expected to begin enrolling in the Fall of 2020.
The award, administered by the National Institute of Arthritis and Musculoskeletal and Skin Diseases, ranks as one of the most significant in SPRIs history, both in size and recognition. Given the potential of regenerative medicine to enhance human health and treat disease, the United States Congress included a provision in the 21st Century Cures Act a law passed in December 2016 to accelerate medical discovery and innovation to support the NIH-established Regenerative Medicine Innovation Project (RMIP). The RMIP aims to accelerate the field by supporting clinical research on adult stem cells while promoting the highest standards for protecting patient safety during the conduct of research.
This is a really great honor for SPRI, said Dr. Huard, who first came to Vail in 2015 and has served as the director of the Center for Regenerative Sports Medicine in addition to his role as the institutes chief scientific officer. Past recipients of these RMIP awards have been Albert Einstein College of Medicine, Boston Childrens Hospital, Columbia University Health Sciences, Childrens Hospital of Philadelphia, Harvard University, University of Colorado Denver and Yale University.So, we are in very good company.
The grant anticipates over $2.8 million from the NIH and requires a 1:1 match from SPRI over the next five years, pending availability of federal funds. The clinical trials and resulting publications and reports will take place over the next five years. A generous SPRI benefactor committed to fund the first year of the match, and Dr. Huard is hopeful that with the NIH matching the funds, more philanthropists will be inspired to become involved in this groundbreaking project.

The trial is entitled,The Use of Senolytic and Anti-Fibrotic Agents to Improve the Beneficial Effect of Bone Marrow Stem Cells for Osteoarthritis. Huard explains in laypersons terms:
The idea behind the trial is to delay osteoarthritis in the knee, said Huard. Our goal is to delay the need for that first knee replacement in a patient for as long as we can. Over time SPRI intends to expand this area of research to other joints including hip and shoulder.
This clinical trial is designed to determine whether senolytic and/or antifibrotic agents will improve the beneficial effect of Bone Marrow Stem Cells for the treatment of symptomatic knee osteoarthritis. The trial will include four groups, totaling 100 patients, to investigate the teams hypothesis that the use of these agents will improve patient outcomes.
One of the great things that I love about this particular clinical trial is that we are actively involving our orthopaedic surgeons and our biomotion lab staff as well, said Dr. Huard. This will truly be a team effort over the next five years.
Those world-class surgeons are led by Dr. Philippon, considered one of the worlds foremost orthopaedic surgeons. The biomotion lab is under the direction of Dr. Tashman. The contributions of these two leaders and the talented roster of surgeons, clinicians and technicians in their departments will be critical to the success of the upcoming clinical trials. SPRIs Center for Outcomes-Based Orthopaedic Research and its director Grant Dornan are also participating in this project by contributing the statistical outcomes.
Dr. Philippon is not only a world-class surgeon but he is also an innovator, added Dr. Huard. He always wants to improve and is still willing to try new things to enhance patient outcomes. Dr. Tashman is the same way. Like everyone here at SPRI and The Steadman Clinic, they are embracing the cutting-edge technology available to them and finding new and better ways to treat patients and, most importantly, reduce patients recovery time and get them back to their active lives as quickly and safely as possible.
Huard notes that the rare combination of a globally recognized research institute like SPRI and a world-class orthopaedic surgery clinic like The Steadman Clinic in the same building is one of the key factors in the awarding of this RMIP grant.

By the time this column is published, the one-year anniversary of my lifesaving stem cell transplant will have come and gone. But as I sit here sweating away in the thick humidity of a rare August heat wave, and as the sun sets upon my little garden in the boroughs of North London, I cant help but bask in this moment of reflection and humble appreciation.
An entire year has come and gone, just like that. Time is such a strange, bizarre phenomenon. It feels as if it has passed by in a blur a simple snap of my fingers and yet so much has transpired. So much has unfurled and unfolded, fallen apart and come together, all in just 12 tiny months.
One year ago today, I was lying in a sterile hospital bed with a drip attached to my veins. I was puffy, exhausted, and frail from the chemicals seeping into my blood. I had no immune system and was days away from having my sisters stem cells infused into my bloodstream, where they would collectively begin to regenerate an entirely new immune system.
A year ago, I could barely walk because I was so weak. I had no connection to my body other than distrust. I was small, sad, tired, and full of grief and heartache. A year ago, the world was painfully heavy and dark. Now, as I sit here staring at the trees in my backyard and watching the sun pour through the gaps in the branches, painting the leaves a brilliant verdant green, I see so much light in the world its almost blinding.
I just returned from a weekend in the English countryside with some of my best friends. We stayed in a house beside the sea on the coast of Cornwall. The sun was warm, and the world felt like it was washed in a soft, tangerine glow. I cant remember the last time I have laughed so much. I felt wonderfully alive, radiating with joy at the sheer beauty of all the small, simple things. The tiny white and yellow daisies we picked from the bushes on the side of the road, the perfect flapping of a butterflys delicate wings, the sound of my friends laughter, the lightness of being, the magnificence of loving, and being loved in return.
This anniversary is deeply significant to me because it symbolizes just how far I have come. If only a year ago, lying in that hospital bed, Id had a crystal ball and could have seen into the future and understood that everything really would be OK. If only I could have seen my strong, able body, my mop of unruly blond curls, my happy, overflowing heart. Back then, it felt like my life was falling apart. Now, despite all the unexpected twists and turns and challenges of this tumultuous year, the disparity between where I was then and where I am now is not lost on me.
I think thats the greatest gift Ive gained from the long periods of sickness and convalescence Ive endured understanding the true value of being healthy.
Nothing can take that lesson from me now, and for that, Im deeply grateful. Ill always be able to look back and recall exactly what it felt like to lie in that ward, watching the clock, wishing time would pass, desperately longing to feel better, to just feel like myself again.
Health and time are the two things we take for granted the most, yet they are the greatest gifts we will ever have.
Despite how quickly this year has passed, it also has been filled with nothing but time. Time to sit and reflect. Time inside. Time alone. Time apart from loved ones. Its been tough, painful, and full of missing. But its also been deeply healing. I have watched as my body has transformed from a weak, thin, pale, depleted shell of skin and bones to a strong, tanned, healthy, muscular machine. I have watched the pigment return to my skin, the smile return to my cheeks, the sparkle return to my eyes. I never could have imagined the progress I would make in a year, and Im so deeply proud of myself for persevering through those dark, drawn-out days.
For anyone facing the eye of the storm now, or fresh off the boat of treatment, I want you to know that it does get better. If only I could whisper in the ear of that scared and sick girl and tell her that everything will be OK. That things wont stay dark forever. To hold on, theres so much more to come.

Beauty companies face an uptick in alleged false-labeling class actions. Whether the actions are justified or vexatious, one thing is certain: they are expensive to defend. By keeping the following labeling-related litigation trends in mind when considering and reviewing product labels and marketing, beauty companies can, hopefully, avoid becoming a litigation target.
All-Natural Claims
This most common type of false-labeling related lawsuit targets products that are labeled as containing a natural ingredient or as being all-natural. Targeted companies and claims have included:
The attorneys who seem to most often bring these actions also have filed lawsuits against food and beverage companies based on the same or similar label claims. The firms are leveraging their experience litigating against food and beverage to pursue the beauty industry.
Beauty companies should bear in mind that on Nov. 8, 2019, HR 5017 was introduced in the House of Representatives. This bill would amend the Federal Food, Drug and Cosmetic Act to define natural with a certain set of standards. As currently drafted, the bills definition of natural identifies a variety of characteristics that the product must have, for instance, it would have to contain at least 70% natural substances and cannot be made using any of seven processes, such as ionizing radiation. There is no present timeline for a vote on the bill, but companies should assume that its adoption will stoke litigation in this category.
Claims v. Labels
For a few thousand dollars or less, anyone can have a product lab-tested to learn whether it contains the ingredients or attributes claimed on the product label. The cases teach a variety of lessons for both labeling and playing class action defense.
In Forouzesh v. CVS Pharm., Inc., Case No. 2:18-CV-04090 (C.D. Cal), the plaintiff purchased a bottle of CVS Sport SPF 100+ Sunscreen Spray, but testing showed that the sunscreen only had an SPF of 29. CVS argued that the plaintiffs claims were preempted (in other words, that the plaintiff sought to impose on CVS a requirement that is different from or in addition to, or that is otherwise not identical with a requirement contained in the FDCA).
Ultimately, the court dismissed the lawsuit, finding the consumer did not adequately show that the product he purchased was in fact the one that his counsel tested, and, the testing methodology did not appear to comply with FDA regulations. The court gave the consumer an opportunity to amend to show such compliance, but the consumer did not timely file an amended complaint.
Sunscreens claiming to be mineral-based are also a target. For instance, in Prescott v. Bayer Healthcare et al., Case No. 5:20-cv-00102 (N.D. Cal.), plaintiffs claim that, Defendants are exposing babies and children to harmful chemical-based ingredients hidden in their sunscreens by fraudulently passing them off as safe mineral-based ingredients.
Aloe vera has also been targeted. In Kalajian v. Rite Aid Corp., Case No. 2:17-cv-06777 (C.D. Cal.), the consumer alleged that according to independent lab tests, the aloe vera products had no actual aloe vera at all. In a parallel case filed in the Northern District of Illinois, consumers argued that there was not enough of aloes active ingredient (acemannan). In this latter case, following several years of litigation, the court found, there is no evidence that a certain amount of acemannan must be present in the finished product for it to be aloe.
Companies intending to infuse their products with CBD should be aware that CBD-related class action litigation has been red hot. Actions have been filed in multiple states with consumers claiming that the CBD content listed on product labels and packaging does not match the actual CBD content. Other CBD-related class actions attack label claims stating that CBD will cure, mitigate, treat or prevent disease, or is intended to affect the structure or function of the body, in violation of FDA regulations. One topical cream has already been targeted. Dasilva v. Infinite Product Co. LLC, Case No. 2:19-cv-10148 (C.D. Cal.) (freezing point cream alleged to freeze away all aches and pains, and an afterglow healing oil alleged to be great for new tattoos, eczema, acne, scarring or open wounds.).
These cases serve as a reminder that, for companies using a marketing amount of an ingredient which requires a higher therapeutic amount to be efficacious, some thought should be given about positioning that ingredient in labeling statements and marketing materials.
Buyers Remorse
The idea of caveat emptor doesnt hold up in a courtroom when it comes to personal care product performance. Consumers have asserted that a wide range of products have not performed as expected (and therefore are falsely labeled), including:
Cases in this category are unique to their facts, making it hard to state a uniform defense to them. Legal departments should coordinate with their product managers and marketers to understand why certain label claims are made to ensure they are defensible.
Undisclosed Side Effects
This trend encompasses products that have caused actual alleged harm and those which are alleged to be dangerous but which may not have caused identifiable physical harm.
In the former category is a case against Rodan & Fields for its Lash Boost product. Lewis v. Rodan & Fields, Case No. 18-cv-02248 (N.D. Cal.). Consumers alleged that the Lash Boost eye serum had harmful side effects linked to the ingredient isopropyl closprostenate (ICP). ICP is a type of synthetic prostaglandin analog, a class of drugs used to manage glaucoma. The consumers alleged a variety of harms which were known side effects of this class of drugs, including changes in eye color, lashes falling out and not growing back, and one plaintiff had her vision damaged. While printed warnings appeared on the outside of the product container and inside on an insert, none referred to the side effects associated with ICP. The complaint referred the court to a warning letter that FDA sent to another maker of cosmetic lash enhancement products that used ICP without including warnings about the possible adverse effects associated with ICP. Based on these allegations, the court found the consumers plausibly alleged violations of five states false advertising laws.
Other cases are based on bare allegations that product ingredients are harmful without parallel allegations of personal injury (only economic damages are asserted). For instance, in Clark v. Kendo Holdings Inc., Case No. CGC-17-562492 (S.F. Super. Ct.), the consumer alleged that certain Ole Henriksen cosmetic products that contained alpha hydroxy acid (AHA) were falsely labeled. AHA in cosmetic products is said to remove the outermost layer of dead skin cells, which produces a short-term cosmetic benefit, but can also increase the damage sun-exposed skin suffers from ultraviolet radiation. The consumer alleged that this causes permanent damage and increases the risk of cancer. He also alleged that the FDA recommends that manufacturers using AHA label their products with a specific sunburn alert, and the products in question did not contain the recommended warning. The parties reached a settlement prior to class certification.
These cases are similar in that the plaintiffs attorneys expressly pointed the courts to FDA enforcement actions or advice (not taken) to bolster the complaints. As described in this article, the same has been done with FTC warnings. The attorneys are plainly using FDA and FTC guidance to guide their litigation strategy. Beauty companies legal departments should stay abreast of FDA and FTC enforcement actions and advice, as these could be harbingers of litigation trends.
Inaccessible Product
If youve ever used a cosmetic product down to the last drop, then youve experienced the tipping and tapping involved in trying to get that last bit of product out of the container. This common consumer experience has proven useful in defeating this final false-labeling class action trend.
In Ebner v. Fresh, Inc., 838 F.3d 958, 965-66 (9th Cir. 2016), the plaintiff complained that only a portion of Freshs Sugar lip product was accessible. Allegedly, the tubes screw mechanism permits only 75% of the total lip product to advance past the top of the tube. A plastic stop device prevents the remaining 25% of the product from advancing, preventing direct application from the tube to the lips. The Ninth Circuit affirmed dismissal of this lawsuit, finding that, [d]ispenser tubes that use a screw mechanism to push up a solid bullet of lip product are commonplace in the market. The reasonable consumer understands the general mechanics of these dispenser tubes and further understands that some product may be left in the tube to anchor the bullet in place. Critically, the label disclosed the correct weight of included lip product.
Despite the favorable defense outcome, plaintiffs have not abandoned this litigation theory. Subsequent lawsuits have targeted a medicated lip balm dispensed through a flexible tube, and a purveyor of cosmetics whose products are sold in sealed bottles, often made of glass, dispensed with a pump.
These cases teach two important defense lessons:
Slack fill lawsuits are a related litigation concept. Slack fill is the difference between the actual capacity of a container and the volume of product contained therein. Some slack fill has a legitimate purpose in product packaging, but nonfunctional slack fill is impermissible. If a cosmetics (opaque) packaging suggests that there is more product inside than there actually is, a beauty company may find itself facing a slack fill class action.
To conclude, by watching trends in related spaces such as food and beverage, noting FTC and FDA enforcement actions against industry peers, and revisiting product labeling with product managers and the marketing team, beauty companies can help to set themselves up to minimize the risk of litigation.

Some skin, eggs and tissue samples are all that remain of Malaysia's last rhino, Iman, who died last November after years of failed breeding attempts.
Now scientists are pinning their hopes on experimental stem cell technology to bring back the Malaysian variant of the Sumatran rhinoceros, making use of cells from Iman and two other dead rhinos.
"I'm very confident," molecular biologist Muhammad Lokman Md Isa told Reuters in his laboratory at the International Islamic University of Malaysia.
"If everything is functioning, works well and everybody supports us, it's not impossible."
The smallest among the world's rhinos, the Sumatran species was declared extinct in the wild in Malaysia in 2015. Once it had roamed across Asia, but hunting and forest clearance reduced its numbers to just 80 in neighboringIndonesia.
Iman, 25, died in a nature reserve on Borneo island, following massive blood loss caused by uterine tumors, within six months of the death of Malaysia's last male rhino, Tam.
Efforts to get the two to breed had not worked.
"He was the equivalent of a 70-year-old man, so of course you don't expect the sperm to be all that good," said John Payne of the Borneo Rhino Alliance (BORA), who has campaigned for about four decades to save Malaysia's rhinos.
"It was obvious that, to increase the chances of success, one should get sperm and eggs from the rhinos inIndonesia. But right till today,Indonesiais still not keen on this."

And even if it worked, the animals' lack of genetic diversity could pose a threat to long-term survival, Galli told Reuters.
Indonesian scientist Arief Boediono is among those helping in Malaysia, hoping success will provide lessons to help his country's rhinos.
"It may take five, 10, 20 years, I don't know," Arief added. "But there has already been some success involving lab rats in Japan, so that means there is a chance."
Japanese researchers have grown teeth and organs such as pancreas and kidneys using embryonic stem cells from rats and mice in efforts to grow replacement human organs.
For now, however, Iman's hide will be stuffed and put on display alongside Tam in a Borneo museum.

A common skincare mistake many people make?
In my opinion, its using toner. Thats a misconception because still, many people use a toner as the second step of cleansing as opposed to the first step of treating the skin, and this is from my experience of talking with clients.
They put toner or micellar water on a cotton pad and they keep wiping and seeing more make-up. If you see more make-up on your cotton pad, that means you need to go back into washing.
Toner is very often misunderstood or skipped, and it shouldnt be. I cant imagine, for instance, applying a serum on my face without applying toner first. There is no way the efficacy of the product will be the same if you have not applied a toner. Depending on the toner, they offer hydration and sometimes micro-exfoliation, but mainly they are used to maintain the pH of the skin. The optimal pH for our skin is 5.5, and many factors from our diet to lifestyle, and even washing our face can throw the skins pH off the scale, so it's very important to balance it back.

With face masks becoming part of everyday life, maskne has become a real problem. How can we prevent these breakouts?
When you wear a mask, it creates a micro-climate and we keep breathing carbon dioxide back and forth, so there is not enough of anti-bacterial oxygen getting into the skin. There is sometimes too much moisture happening, so we will get super hydrated initially, and then get quite dehydrated right after. Thats when you will experience eczema and redness.
What I recommend is keeping the skin as clean as possible before wearing the mask, with just a balancing toner, and protecting balm or healing ointments to lubricate areas where the mask could potentially irritate the skin. Very often, its on the nose bridge, as well as on the side and behind the ears.
For less reactivity, I wouldnt go through with the whole routine, but if you have to, I would advise starting your routine earlier so the products are on your skin for at least 30-40 minutes. If you will be stepping out shortly, reduce the routine and skip some steps. But no matter what, never forget about your SPF because the friction of the mask could also get rid of our stratum corneum and create little scabs, causing discolourations.
Then, as soon as you arrive home, take the mask off, wash your face, and again balance your skin with toner and use your serums.
What are your tips for soothing breakouts or eczema caused by wearing masks?
Even with microscopic breakouts, I would continue using any product that is hydrating because sometimes we misunderstand we have a breakout and then we try to use benzoyl peroxide, or everything that is dehydrating. No, your skin would be producing even more sebum. So keep hydrating your skin with soothing ingredients like colostrum and hyaluronic acid.
Your skincare routine?
My morning routine is very brief: cleanser, toner, a serum and then there is moisturiser. For my night time routine, my very first step is getting into the shower when I get home. I begin with massaging my body with my shower gel and silicone gloves under the shower, then I apply products like multi-vitamin oils and sometimes micro-exfoliating toners all over my body.
Then, I go to my face. I usually dont wear any make-up, so I start right away with my cleanser with some massaging movements and I remove it with a linen washcloth, followed by a toner. My favourite way of applying toner is the sponge techniqueinitially, you spread the product on your face, and then you press and release. When you press, your skin microscopically opens and when you release, the skin grasps whatever is on the surface.
After the toner, I use my serum. Ive been using the Capture Totale C.E.L.L. Energy Super Potent Serum since September. If you have weaker areas like forehead lines and nasolabial folds, these are the areas I would concentrate longer on, followed by an eye cream and moisturiser. Thats usually my five-step basic routine.
About twice a week, I do facial masks, one of those quick ones because Im the kind of New Yorker who only has five seconds for myself. New Study Presents Cell-based Therapy for MN Diseases or Spinal Cord Disorders – Mirage News
The spinal cord is a bundle of nerves inside the spine that gives your body structure and support. Spinal cord injuries (SCIs) tend to be devastating and most are permanent. Recent research has shown that motor neuron obtained from skin cells could serve as potential treatments for spinal cord injuries, and thus has received considerable research attention. With this, a new door has been opened for treating not only spinal cord injuries, caused by workplace accidents and car crashes, but also Lou Gehrigs disease, known as amyotrophic lateral sclerosis or ALS.
A research team, led by Professor Jeong Beom Kim and his research team in the School of Life Sciences at UNIST has demonstrated that human fibroblasts can be converted into induced motor neurons (iMNs) by sequentially inducing two transcription factors, POU5F1(OCT4) and LHX3. The research team further investigated the therapeutic effects of iMNs for treating traumatic spinal cord injury using rodent spinal cord injury model. Their findings indicate that the sequential induction of two transcription factors is essential for generating self-renewing iMNICs more efficiently. This method not only ensures large-scale production of pure iMNs, but also facilitates the feasibility of iMNs for SCI treatment.
The spinal cord is responsible for transmitting signals from the brain to the rest of the body, and vice versa. Along with motor and sensory deficits, damage to the spinal cord can cause long-term complications, including limited mobility. Although there are many treatment options available for people with SCI, most of them have adverse side effects that impact therapy. And this is why stem cell (SC) therapies to restore functions of damaged tissues are attracting attention, recently. Among those cells constituting the spinal cord, motor neurons that involved in the regulation of muscle function have emerged as a promising candidate for the stem cell-based therapy for SCIs. Despite these encouraging advances, ethical issue of embryonic stem cells (ESCs) and tumorigenic potential of induced pluripotent stem cells (iPSCs) have impeded their translations into clinical trials.

To overcome these limitations, Professor Kim and his research team established an advanced direct conversion strategy to generate iMNs from human fibroblasts in large-scale with high purity, thereby providing a cell source for the treatment of SCI. These iMNs possessed spinal cord motor neuronal identity and exhibit hallmarks of spinal MNs, such as neuromuscular junction formation capacity and electrophysiological properties in vitro. Importantly, their findings also show that transplantation of iMNs improved locomotor function in rodent SCI model without tumor formation. According to the research team, This proof-of-concept study shows that our functional iMNs can be employed to cell-based therapy as an autologous cell source. Through this, they resolved the problem of immune rejection, and thus reduce the risk of cancer.
In the study, we succeeded in generating iMNs from human fibroblasts by overexpressing POU5F1(OCT4) and LHX3, says Hyunah Lee (Combined MS/Ph.D program of Life Sciences, UNIST), the first author of the study.

The developed motor nerve cell manufacturing method has the advantage of being capable of mass production. A sufficient amount of cells is required for patient clinical treatment, but the existing direct differentiation technique has limited the number of cells that can be obtained. On the other hand, the method developed by the research team is capable of mass production because it undergoes an intermediate cell stage capable of self-renewal. After injecting the produced cells into the spinal cord injury mice, it was confirmed that the lost motor function is restored and the nerves are regenerated in the damaged spinal cord tissue.
Although further investigation on mechanism responsible for cell fate conversion may be needed, our strategy is a safer and simpler methodology that may provide new insights to develop personalized stem cell therapy and drug screening for MN diseases or spinal cord disorders, says Professor Kim. If combined with SuPine Patch, an adhesive hydrogel patches with the purpose of regenerating the damaged spinal cords, its therapeutic effects will be maximized. He adds, As the incidence of spinal cord injury is high due to industrial accidents, synergistic effects with public hospitals specializing in industrial accidents scheduled to be built in Ulsan should be expected.
This study has been jointly carried out with Professor Kims startup company, SuPine Therapeutics Inc. with the support of the Ministry of SMEs and Startups (MSS). The findings of this research have been published in the 2020 June issue of the online edition of eLife, a renowned academic journal of the European Molecular Biology Organizationl (EMBO).
